"The weather is finally cooperating, so it is now time to fly," said Launch Director Pete Nickolenko.
"Endeavour has patiently waited for this," replied Endeavour commnader Mark Polansky. "We're ready to go, and we're going to take all of you with us on a great mission."
The countdown is about to pick up from a hold at T minus nine minutes - a mark not reached in each of Endeavour's five previous launch attempts since June 13.
An updated forecast recently gave a better than 90-percent chance of weather cooperating.
Barring any late technical issues, Endeavour's three main engines will ignite 6.6 seconds before liftoff, in what amounts to a test firing.
Twin solid rocket boosters will ignite at T minus zero, when giant bolts holding the shuttle to its mobile launcher platform should explode and release the vehicle for a nine-minute climb to orbit.
The boosters separate just over two minutes into flight.
